Synopsis "Methodenentwicklung und Validierung für die Voriconazol-Bestimmung (in German)"

Für die Bestimmung von Voriconazol wurde eine schnelle, empfindliche und spezifische RP-HPLC-Methode mit UV-Detektion entwickelt und validiert. Die Chromatographie wurde auf einer Enable C18 G-Säule (250 × 4,6 mm) im isokratischen Modus unter Verwendung einer mobilen Phase aus Methanol und Acetonitril (60:40, v/v) durchgeführt. Die Durchflussrate wurde auf 1 mL/min eingestellt, und der Abfluss wurde bei 223 nm überwacht. Die chromatographische Analyse ergab einen Hauptpeak für Voriconazol bei einer Retentionszeit von 2,841 Minuten. Als Lösungsmittel für die Analyse wurde Methanol gewählt, wobei die λmax bei 223 nm bestimmt wurde. Die Methode zeigte Linearität über einen Konzentrationsbereich von 5-30 g/ml. Die Validierungsparameter, einschlie lich Linearität, Bereich, Präzision, Genauigkeit, Nachweisgrenze (LOD) und Bestimmungsgrenze (LOQ), wurden bewertet und entsprachen den erforderlichen Standards.
